چکیده

Bull’s fertility is highly important as it involves the fertilization of oocyte to produce a good, viable and genetically potential conceptus. Artificial insemination is the most widely used technique followed throughout the world, to increase numbers of genetically potential breeding bulls, offering many advantage over natural service. High quality semen is crucial for successful artificial insemination. Though the technique is being used to increase bull fertility status still the numbers of efficient breeding bulls are reduced day by day. Male related embryonic death owe to reduced semen quality due to heat stress (Setchell et al., 1988; Colas, 1983). Heat stress affects the body growth, the biological functions and the productive and reproductive characteristics of sheep (Sevi et al., 2002). Season is one of the major factors influencing the reproductive performance of breeding bulls and it exerts its effect through macro and micro climatic factors like temperature, humidity, rainfall, photo-period (Mandal et al., 2000). In case of bulls the increased evidence of sperm abnormality and reduced sperm output has been reported during warmer seasons of the year (Igboeli et al., 1971; Parkinson, 1987). In the light of above facts, it is very essential to evaluate the semen quality before artificial insemination. Semen quality study can be best assessed by the funtional integrity of sperm membrane in cattle (Correa et al., 1994). So the present study was undertaken with the objective to find out the status of membrane integrity of fresh and post thaw sperm in bulls during different range of temperature

Cadmium as an etiology of sperm dysfunction in Holstein bulls

Oxidative stress has been identified as a crucial factor leading to male infertility largely due to peroxidativedamage to the sperm cell membrane. Cadmium (Cd) is a widespread environmental pollutant and oneof the well-known reproductive toxicants. Because of its long biological half-life (10–30 years), Cdaccumulates in the biological systems. The present study was designed to assess the concen...

P-137: Effect of Different Equilibration, Freezing and Thawing Rate on Cryopreserved Buffalo Spermatozoa Diluted inTris- Egg Yolk Extender

Background: Artificial insemination (AI) has been used for increasing genetic potential of animals in production while application of AI has been restricted in buffalo due to low freezing ability and fertility of frozen-thawed buffalo spermatozoa. So improvement in buffalo sperm cryopreservation may enhance the breeding programs of this valuable farm animal. Semen processing is one of the major...

P-138: Effect of Short Term Storage on Functional Parameters of Post-Thawed Buffalo Spermatozoa

Background: Detection of sperm parameters based on conventional methods, immediately after thawing, have been revealed sperm capacity to in vivo and in vitro fertilization. But this evaluation is not fully reliable because of long journey of sperm during female genital tract to reach the oocyte and fertilize it. Therefore, analyzing postthawed sperm parameters after incubation is more beneficia...

P-73: Effect of Sodium Dodecyl Sulfate and Different Levels of Clove Bud Extract on Motility, Viability and Plasma Membrane Integrity of Ram Spermatozoa during Cooling and Cryopreservation

Background: Hen egg yolk is a common component of most semen cryopreservation extenders. However, diluents containing the egg yolk may have adverse effects on the quality of the semen. Egg yolk could be solubilized by the addition of sodium dodecyl sulphate (SDS) in semen extenders. Many studies have shown beneficial effects of antioxidants addition to semen freezing extenders. Antioxidants are...

O-12: Studies on Sequestration of PDC-109 Protein on Cryodamage and In Vitro Fertility of Crossbred Bull Spermatozoa

Background: Plasma membrane of the spermatozoa interacts with and is altered by abundant seminal PDC-109 present in their immediate milieu in vivo with damaging effect in a time- and concentration- dependent manner. Therefore we hypothesized that sequestration of detrimental protein from ejaculates would be beneficial for cryopreservation of sperm cells. To this aim we evaluated the effect of s...
